
Advancements in Single-cell CRISPR Screens Enhance T Cell Immunotherapy
Researchers have used single-cell CRISPR screening to map the gene regulatory networks that control the fate of T cells in cancer. By targeting transcription factors (TFs) in CD8+ T cells and analyzing their effects on gene expression, the study identified co-functional modules and gene programs associated with effector function, exhaustion, stemness, and proliferation. The study also revealed the IKAROS-TCF-1 axis as a key regulator of the transition from exhausted T cells to effector T cells. These findings provide insights into the molecular mechanisms underlying T cell differentiation in the tumor microenvironment and could inform the development of new immunotherapies.
