Using single-nucleosome tracking and super-resolution imaging in living human cells, researchers show that cohesin constrains condensed euchromatin domains by loop formation, preserving transcriptional insulation; cohesin loss increases nucleosome mobility and local domain intermixing without majorly changing overall compaction, and Hi-C data indicate domain mixing at short scales with increased long-range contacts.
