High-Dose Vitamin D3 Fails to Extend Progression-Free Survival in Untreated Metastatic Colorectal Cancer

In the SOLARIS phase 3 trial, 455 patients with previously untreated metastatic colorectal cancer were randomized to high-dose vitamin D3 plus standard chemotherapy with bevacizumab or standard-dose vitamin D3. High-dose vitamin D3 did not improve progression-free survival (11.8 vs 10.3 months), objective response rate, or overall survival, though there was a signal suggesting left-sided tumors may benefit. The regimen was well tolerated with similar adverse events between groups. The findings do not support routine use of high-dose vitamin D3 in unselected patients and warrant further investigation in targeted subgroups.

Zynlonta safety concerns rise as LOTIS-5 shows higher deaths

Merck's WELIREG® Phase 3 Trial Shows Promising Results in Advanced Renal Cell Carcinoma

Merck has announced positive Phase 3 trial results for WELIREG (belzutifan), its oral hypoxia-inducible factor-2 alpha (HIF-2α) inhibitor, in the treatment of advanced renal cell carcinoma (RCC) that has progressed following PD-1/L1 checkpoint inhibitor and vascular endothelial growth factor-tyrosine kinase inhibitor (VEGF-TKI) therapies. The trial showed statistically significant improvements in progression-free survival (PFS) compared to everolimus, as well as a significant improvement in objective response rate (ORR). While there was a trend towards improvement in overall survival (OS), it did not reach statistical significance. The safety profile of WELIREG was consistent with previous studies.
