"Exploring the Efficacy of Intravenous Vitamin C for COVID-19 Hospitalized Patients"

Two prospectively harmonized randomized clinical trials, involving 2,590 patients hospitalized with COVID-19, investigated the efficacy of intravenous vitamin C in improving organ support-free days and hospital survival. The trials found that vitamin C had a low probability of improving the primary composite outcome of organ support-free days and hospital survival in both critically ill patients and those who were not critically ill. The posterior probabilities for efficacy were 8.6% among critically ill patients and 2.9% among non-critically ill patients. The trials were terminated early due to statistical triggers for harm and futility being met.

"Promising immune modulation strategy launched for hospitalized covid-19 patients"

A clinical trial has been launched by the National Institutes of Health (NIH) to test the effectiveness of early intensive immune modulation for hospitalized COVID-19 patients with mild illness. The trial, part of the global clinical trials consortium known as STRIVE, aims to enroll approximately 1,500 participants at research sites worldwide. The study will investigate whether adding a second therapeutic earlier in the patient's treatment can prevent progression to severe illness and improve recovery. The drug being tested is abatacept, which is believed to be useful as an immune modulator for COVID-19 by suppressing overactive immune responses. The trial will monitor patient safety and assess the benefits of early immune modulator intensification.

The Impact of the Pandemic on Hospital Outcomes and Healthcare Disruptions

A retrospective study examining hospitalizations in Canada found that outcomes for hospitalized patients without COVID-19 did not differ significantly before and during the pandemic, except for specific subgroups such as those with heart failure or COPD/asthma. Patients with non-COVID conditions and COVID-19 co-infection had a higher 30-day mortality rate and longer hospital stays compared to those without COVID-19. The study suggests that Canadian hospitals, which were already operating at near-capacity before the pandemic, were able to maintain stability in outcomes for non-COVID patients despite the surge in COVID-19 cases. Further research is needed to understand if these findings hold true in other countries.

Anti-inflammatory Drugs Prevent COVID-19 Deaths, Study Shows

A clinical trial studying the use of crizanlizumab, a P-selectin-blocking agent, in hospitalized patients with COVID-19 found that it did not affect mortality or the number of days free of respiratory or cardiovascular organ-support measures. The trial was terminated early due to futility. The results suggest that inhibition of P-selectin is unlikely to benefit patients with moderate-to-severe COVID-19.
